Notes:
Temperature : 27 to 28℃, non-humidified , regular atmosphere (do not use CO2). Use T-flask , shake flask or spinner bottle with proper gas exchange.

Seed cells at 4 × 105 viable cells/mL in sterile environment containing pre-warmed I-MAX medium.

Subculture cells when cell density reaches 2 × 106cells/mL, normally twice a week.

Sf-9 and Sf-21 cells are not anchorage dependent. They can grow in monolayer or suspension. Insect cells are sensitive to physical shearing so culture conditions must be carefully established.

Adaptation Of Cells To IMAX Serum-free Media :

It is important that cell viability is 90% and in the mid-logarithmic phase when adapting from serum containing medium to I-MAX medium.

For monolayer cultures simply exchange the culture medium with prewarmed (27℃) I-MAX. For suspension cultures centrifuge the cells, aspirate the medium and add prewarmed I-MAX at a cell density of 5 × 10⁵ cells/mL.

In the rare cases when direct adaptation does not work use sequential adaptation. Culture cells into increasing ratios of I-MAX by first using a mixture of 25% I-MAX and serum containing medium, followed by a mixture of 50%, 75% and 100%.
