CHO-200 serum-free medium is designed for the growth of various Chinese hamster ovary (CHO) cell lines, including transfected cells. Optimized specifically for protein production, CHO-200 contains advanced additives that effectively promote protein expression while moderately slowing the cell growth rate. This medium is formulated for use in suspension culture production systems.

Instructions For Use:
Adaptation Of Cells To CHO Serum-free Media :
Direct adaptation:
1. In most cases it is possible to change the cells directly from the serum containing media to serum free media under normal culture conditions.
2. In most cases it is possible to seed CHO cells that have been removed from freezing medium directly in the serum-free medium, when the cell concentration is at least 5 x 105cells per 23cm2. The cells will begin to grow and after a few passages the adaption will be complete.
Importance:
The cell viability should be over 90% and the cells should be in mid-logarithmic phase of growth at the time of serum free media.
Note:
However, there are cases where the cells do not adapt successfully after direct transfer and it is necessary to perform gradual adaptation. The cells should be seeded at 3 x 105per mL in a mix of serum-free medium CHO-200 ( 1:1, V/V ) and serum containing media. The serum containing media is reduced to 25% after 2 passages and then cells are grown in serum free media. In order to save time, we recommend parallel experiments with direct adaption and with weaning. Normally, after the second transfer, it will be obvious whether direct adaption has been successful, and if not, only the weaning experiments are continued.
