Adaptation Of Cells To SFM290 Serum-free Media :
Direct adaptation:
In most cases it is possible to change the cells directly from the serum containing media to serum free media under normal culture conditions. Use your normal established procedures for seeding and growing.
Importance:
The cell viability should be over 90% and the cells should be in mid-logarithmic phase of growth at the time of serum free media.
Note:
However, there are cases where cells do not adapt successfully following direct transfer, making gradual adaptation necessary. Cells should be seeded according to your established procedures in a mixture of serum-free medium SFM 290 (1:1, V/V) and serum-containing medium. The proportion of serum-containing medium is reduced to 25% after 2 passages, after which cells are cultured in serum-free medium. To save time, we recommend performing parallel experiments using both direct adaptation and weaning protocols. Normally, it will become evident after the second transfer whether direct adaptation has been successful; if not, only the weaning experiments are continued. In some cases, supplementation with 1% to 2% FBS is required for continuous culture.
