Product Description:

WISENT HCELL-300, HCELL-400, HCELL-500 products are serum-free alternative for Hybridoma cell culture and high efficiency monoclonal antibodies production, containing a very low protein concentration (under 20 mg/L). Traditionally hybridomas were cultured in media containing serum. Use of serum in culture has many disadvantages and risks for antibody production. The result is a trend towards serum free media for production purposes. The lack of serum requirement ensures consistent culture conditions.

Instructions For Use:

Adaptation Of Cells To HCELL Serum-free Media :

Adaptation will depend on the particular cell line. For some cell lines direct adaptation is possible while others require sequential adaptation. In both cases cells must be used in mid-logarithmic growth phase with high viability.

1) Direct Adaptation

1.Transfer hybridoma cells growing in media containing serum to HCELL medium which has been prewarmed to 37°C. Seeding density should be double the normal seeding density for the particular cell line.

2. Grow cells until viable cell density reaches 1 × 106/mL. Subculture the cells to a cell density of 1 × 105/mL in fresh HCELL medium for 3 passages.

3. If the culture fails to maintain viability over 3 passages, use the sequential adaptation method.

2) Sequential Adaptation

1. Inoculate hybridoma cells at double the normal seeding density in a 75:25 per cent mixture of serum containing media and HCELL.

2. As soon as the culture density reaches 1 × 106 viable cells/mL subculture into a 50:50 per cent mixture of serum containing media and HCELL.

3. As soon as the culture density reaches 1 × 106 viable cells/mL subculture into a 25:75 per cent mixture of serum containing media and HCELL.

4. As soon as the culture density reaches 1 × 106 viable cells/mL subculture into a 100 per cent HCELL media.

FORM: Liquid, Sterile, Ready to use.

SHEIF LIFE: Maximum 18 months.

STORAGE TEMPERATURE: STORE AT 2 TO 8℃. Protect from light.
